Do all Self IP addresses need to be on unique VLANs?
Will configuring 2 unique subnets on the same VLAN cause issues?
For example, if I have 2 unique subnets, do I also need 2 VLANs? One for each subnet.

You'll need two vlans to make everything work properly. Otherwise, layer 2 within your network won't work as you intend it to.

However bigip (Like almost any other IP box) doesn't care if you have multiple subnets on the same VLAN. And you can pretty much ignore layer2 issues for almost anything, as long as the hosts you're using only use IP, and they have the correct network and mask configured (i.e. They'll still use the BigIP as a gateway to talk between the two subnets).
About the only real layer2 issue you should see is that fact that the hosts CAN talk directly to each other by simply configuring the same network subnets (i.e. There's nothing to stop a host on one subnet from talking to the other subnet without going via the bigip if it configures an IP on the second subnet directly).
[i.e. it depend on what you intend layer2 in your network to do. If it's a security thing, don't do it. If it's simply admin (i.e. you ran out of addresses on one subnet), feel free]
