1:should I enable UIE profile in default persist profile,why?
I see we use it via irule,why should enable it in default persist profile
2:should I enable oneconnect profile while using UIE ?
why?
I can see it will send all http request in a keep alive connection to one single backend server without oneconnect,but I am fine with this

I see we use it via irule,why should enable it in default persist profilei do not know but i think it is not different between specifying irule in universal profile and assigning irule to virtual server. anyway, i always assign irule to virtual because not only persistence i do in irule. so, i think it makes more sense to assign it to the virtual.

why?i think you should if you want to reduce a number of connection toward server.

The main advantage to using a UIE persistence profile is that you can share the same profile across virtual servers and use the 'match across' features of the profile.

You can also enable persistence mirroring on the profile but not directly in an iRule.
