We are using F5 for load balancing and we have 2 instances running. If one of the instance is down, will the f5 server know and route all the requests to the second server? Can anyone explain how...

Different Ports means different pools, so you can configure two pools, one for each instance, and configure different health monitors.Both instances will be independent from each other.
shubhamv5_37337The ports are different but in the console I see that both the apps are in same pool. Is it possible to have different health monitors for both of them. Please find the attached image. It looks like both have different health monitors.

You can assign different health monitors for different members within the same pool.
Pool > Pool_Name > Memebers > member_name , switch to Advanced configuration.
Under health monitors choose "Member Specific" , and apply the required monitor.
you may also need to check the "Availability Requirement" option under pool configuration.
