Do i need to use multiple partitions or not?

You can have this set up with out route domain as long as they are routed properly in BIG IP. If you have external Vlan and internal Vlan configured in BIG IP, then you can simply create two partition and create respective VIPs in there.

 

It is up to you if you want to create wide ip for internal VIP.
